PHARMACODYNAMICS
Tramadol is a centrally acting synthetic analgesic compound, which effectiveness at therapeutic doses is due to the synergy:
- of an opioid effect due to fixation on μ opioid receptors,
- of a central monoaminergic effect due to inhibition of reuptake of norepinephrine / noradrenaline and serotonin, acting as a control mechanism of the central nociceptive transmission.
Like other products of this class, Tramadol has antitussive properties. At therapeutic doses, the effect of Tramadol on the gastro-intestinal tract is minimal. The respiratory depressant effects of Tramadol are lower than those of morphine. Animal research has demonstrated a low dependence potential compared to morphine, and a minimal tolerance potential.
